چکیده

Duchenne muscular dystrophy (DMD) is caused by the absence of the protein dystrophin in the muscle cells. The function of dystrophin is still not clear. For enabling study of the molecular function of dystrophin, we used small inhibitory RNA (siRNA) for suppressing the expression of the protein in two muscle cell lines and achieved a quantitative knockdown. We applied two-dimensional differential gel electrophoresis (2-D DIGE) in this new in vitro model for DMD to investigate if the absence of dystrophin during myogenesis causes any changes in protein expression. We did not observe statistical relevant changes. The result of our study suggests that the absence of dystrophin does not have any effect

Drosophila Dystrophin is required for integrity of the musculature

Duchenne muscular dystrophy is caused by mutations in the dystrophin gene and is characterized by progressive muscle wasting. The highly conserved dystrophin gene encodes a number of protein isoforms. The Dystrophin protein is part of a large protein assembly, the Dystrophin glycoprotein complex, which stabilizes the muscle membrane during contraction and acts as a scaffold for signaling molecu...

Inositol 1,4,5-trisphosphate (IP3)-dependent Ca2+ signaling mediates delayed myogenesis in Duchenne muscular dystrophy fetal muscle.

Duchenne muscular dystrophy (DMD) is a progressive neuromuscular disorder characterized by muscle wasting and premature death. The defective gene is dystrophin, a structural protein, absence of which causes membrane fragility and myofiber necrosis. Several lines of evidence showed that in adult DMD patients dystrophin is involved in signaling pathways that regulate calcium homeostasis and diffe...

HSPB7 interacts with dimerized FLNC and its absence results in progressive myopathy in skeletal muscles

HSPB7 belongs to the small heat-shock protein (sHSP) family, and its expression is restricted to cardiac and skeletal muscles from embryonic stages to adulthood. Here, we found that skeletal-muscle-specific ablation of the HspB7 does not affect myogenesis during embryonic stages to postnatal day 1 (P1), but causes subsequent postnatal death owing to a respiration defect, with progressive myopat...

Modulation of Stem Cells Differentiation and Myostatin as an approach to Counteract fibrosis in Muscle Dystrophy and Regeneration after Injury PRINCIPAL INVESTIGATOR:

The loss of myofibers in Duchenne muscular dystrophy (DMD) caused by the absence of dystrophin is aggravated by lipofibrotic degeneration. A promising therapeutic strategy is based on implanting dystrophin + muscle derived stem cells (MDSC) engineered or modulated to commit into myogenic rather than lipofibrogenic lineages. Inhibiting or counteracting myostatin (a negative regulator of muscle m...

Improved regenerative myogenesis and muscular dystrophy in mice lacking Mkp5.

Duchenne muscular dystrophy (DMD) is a degenerative skeletal muscle disease caused by mutations in dystrophin. The degree of functional deterioration in muscle stem cells determines the severity of DMD. The mitogen-activated protein kinases (MAPKs), which are inactivated by MAPK phosphatases (MKPs), represent a central signaling node in the regulation of muscle stem cell function. Here we show ...
